Hey guys, new to the site. I have a question about converting a dual-sport 450 to a supermoto. My question is: will throwing a 320mm front rotor fit in the stock caliper? Or do I need a whole new brake set-up? Thanks!

It's entirely up to you.

What budget are you on?
What use?
Personal preference.

I've raced a KTM with just a bigger disc and relocation bracket. Seemed fine. (Some of the thicker 320mm discs won't fit in the standard caliper - KTM had a special "thin disc")

I've since upgraded to a whole new brake $ystem. I like it better.

If you have a good budget, are a heavy braker, race high speed circuits, or want the best, consider something like a Motomaster 4 piston caliper, heavy duty ("flame") disc and matching radial caliper.
Or
A Berenger 310mm set up - considered by many the "ultimate".

My suggestion : try the disc and relocator first.
